SRX21613503: WGS of VLP isolated from Leishmania infantum

Design: Virus-like particles are isolated from samples by centrifuging culture supernatant and filtering through a 0.2uM filter, with the filtrate concentrated in SM buffer using Amicon-15 centrifugal filter and incubated with DNase I and RNase. Nucleic acid was extracted using AllPrep DNA/RNA Mini Kit. RNA was reverse transcribed into cDNA using a SuperScript III First-Strand Synthesis System Kit.

We isolated viral particles from Leishmania samples and identified viral contigs through metagenomic sequencing, with subsequent detection of these viral contigs in negative controls (culture media and H2O) suggesting that the identified viruses have a laboratory origin.
